The aim of the pilot study was to investigate the acute genoprotective effect of grape seed extract (GSE) on human lymphocytic DNA. After overnight fasting, seven subjects (age 29-60 years) were given 3 mL of venous blood before ingesting 125 mg commercial GSE which was emptied from five capsules. All seven subjects drunk 500 mL of water along with the GSE. Subjects remained fasting over 2.5 hours and another venous blood sample was taken again. The two blood samples, at the same time, underwent comet assay to determine DNA damage after treatment with 50 mM hydrogen peroxide. Control experiment was the same as mentioned above with just 500 mL of water without GSE. All subjects attended both active and control experiments, at least seven days apart, in randomized order.
Results showed that supplementation of five capsules GSE were able to lower cellular DNA damage induced by hydrogen peroxide within 2.5 hours. The study demonstrated that a dose of 125 mg of GSE could be absorbed in blood stream quickly and conferred protection to blood cell DNA.
